چکیده

A study was made of the enzymic degradation of 125I-labelled parathyroid hormone by rat kidney microsomes. Incubation with microsomes resulted in rapid destruction of the labelled hormone. The microsomal factor was not separable by dialysis, and the reaction was favoured by pH values in the physiological range. Velocity of the reaction varied directly as the substrate concentration, and additional crude parathyroid hormone (trichloroacetic acid-precipitated, 3-68mg./ ml.) inhibited destruction of labelled hormone. There was much less inhibition with added trichloroacetic acid-precipitated calcitonin (3.92 mg./ml.) and virtually none with added pig insulin (3.80mg./ml.). Gel filtration of control medium on P6 (Bio-Gel) yielded one radioactive peak at the void volume. After incubation with microsomes three further peaks were obtained on gel filtration. Only the voidvolume peak contained intact 125I-labelled parathyroid hormone, indicating that the microsomal enzyme degraded labelled hormone to a number of smaller fragments.

Measurement of proteolytic activity in biological materials using 125I-labelled casein [proceedings].

The use of radioactively labelled proteins for studying proteolysis is well established (Schneider et al., 1965), as are techniques for radiolabelling proteins, which have been developed mainly for radioimmunassay procedures (Yalow & Berson, 1966). 1311 has been the favoured isotope for such work, although it has a short half-life (8.04 days). "'I has a longer half-life (60 days) and can be eff...

The Labelling of Proteins to High Specific Radioactivities by Conjugation to a 1251 - Containing Acylating Agent APPLICATION TO THE RADIOIMMUNOASSAY

1. A new method is described for labelling proteins to high specific radioactivities with 1251. The protein is treated with a 25I-labelled acylating agent, iodinated 3-(4-hydroxyphenyl)propionic acid N-hydroxysuccinimide ester, which reacts with free amino groups in the protein molecule to attach the 125I-labelled groups by amide bonds. 2. Three protein hormones have been labelled by this metho...

17α-Hydroxy-4-pregenen-3-one (17 αP) assay, using acetylcholinesterase enzyme as tracer

17a-hydroxy-4-pregnen-3-one (17aP) hormone is a precursor of other steroid hormones and radioimmunoassay has already been used to measure it, but a simple and rapid "Enzyme Linked Immunosorbant Assay" (ELISA) is described and validated here. A general procedure for prepara-tion of the acetylcholinesterase labelled steroid is described, which is applicable to any steroid. Use of acetyl...

Comparison of the in situ degradation parameters of fresh and frozen-thawed 15N-labelled alfalfa and ryegrass

Degradation of dry matter (DM) and nitrogen (N) in fresh or frozen-thawed alfalfa and ryegrass was studied by using in situ technique. The forages were labeled with 15N during growth in a glasshouse, harvested at similar growth phase, and fresh (F) or frozen-thawed (FT) samples were incubated in the rumen of 3 sheep. There was no difference (P>0.05) between forage type (S) for the immediately s...
